Effect of Amethopterin on the Hemopoietic Function of the Bone Marrow in Rats

Abstract
In order to evaluate the toxicity of amethopterin on the bone marrow, serial observation of the peripheral blood and bone marrow was done after injection of amethopterin into the rats. The results were as follows.
1. The bone marrow of the rats administered with small doses of amethopterin (0.025mg/l00gm) showed moderate depletion of hemopoietic cells, especially of erythroid cell.s.in the later stage of the first five-day course. At the end of the second five-day course, the depletion was severe. Granulocytes in the peripheral blood decreased significantly in number during the second five-day course.
2. The bone marrow of the rats administered with large doses(0.05mg/l00 gm) showed conspicuous cellular depletion from the initial stage. At the end of the second five-day course, only a few, scattered blast cells and reticular cells were observed. The number of granulocytes in the peripheral blood decreased in concordance with the degree of bone marrow suppression.
From, these results, it was concluded that decrease of the peripheral white blood cells was preceded by bone marrow suppression which caused more marked decrease of the granulocytes.
